Samuel de Assis was born on 5 May 1982 in Aracaju, Sergipe, Brazil, to Mr. and Mrs. Assis. He comes from a family of five children — he is the middle child and the only Black member of his family.

Before pursuing acting, de Assis studied Chemistry in Sergipe and was a professional basketball player, playing as a point guard for the state of Sergipe and for Flamengo. He later moved to São Paulo to study theater and began his acting training at Teatro Oficina.
