Matt Iseman was born on January 22, 1971, in Denver, Colorado, to parents Michael Dee “Mike” Iseman, a pulmonologist, and Joan Christensen. He has one brother, Thomas.

He earned a Bachelor of Arts with honors from Princeton University and a Doctor of Medicine from Columbia University Vagelos College of Physicians and Surgeons. Matt Iseman began his professional life as a physician after completing medical training. While finishing his residency he experienced an existential shift and decided to pursue stand-up comedy full time, remaining a licensed doctor.
